Electron Microscope:

Light Microscopes are restricted by the physics of light to 500x to 1000x magnification and a resolution of 0.2 micrometers. Such restriction made the development of Electron Microscopy. Therefore, the theoretical limit was reached in the early 1930s and there was a technical urge to examine the ultra structure and the good structure of cells.

To notice the fine structure there is a requirement for 10,000 x plus magnification, that is not possible by Light Microscopy. The Electron Microscope was developed in the year 1931 in Germany by Max knoll and Ernst Ruska. The primary scanning electron microscope was developed in 1942 and the Electron Microscopes were made obtainable commercially around the year 1965.

Electron Microscope works identical to that of light microscopes except that they employ a focused beam of electrons rather than light to “image” the specimen

The following basic steps are included in Electron Microscopes.

1. A beam of electron is developed from an electron source and accelerated towards the specimen employing a positive electrical potential.

2. The beam is confined and focused employing metal apertures and magnetic lenses into a thin, focused monochromatic beam.

3. The beam is focused on to the sample utilizing a magnetic lens.

4. Interactions take place within the irradiated sample, affecting the electron beam. Such interactions and effects are detected and converted into an image.

There are two kinds of Electron Microscopes. They are:

1. Transmission Electron Microscope
2. Scanning Electron Microscope
