Replication:

Replication is a process where DNA copies itself to generate identical daughter molecules of DNA.

Models of Replication:

Three models of replication had been planned. They are:

1. Conservative replication
2. Dispersive replication and
3. Semi conservative replication

Conservative replication:

As per to this model, the parental DNA is conserved to one daughter cell and the newly synthesized DNA to other daughter cell.

Dispersive Replication:

As per to this model, the parental DNA is not equally distributed (randomly) to the daughter cells.

Semi conservative Replication:

Semi conservative model was established to be the correct mode of replication via the experiment performed through Messlson and Stahl in the year 1957. E. coli cells were grown in a medium consisting of 15NH4Cl, for several generations like that the E.coli cells comprise density labelled 15N atoms in their DNA. After that the cells were grown in a medium consisting of unlabelled NH4Cl. DNA was harvested from the daughter cells later than the one generation and the density of the DNA was analyzed by using Cesium chloride density gradient centrifugation. If replication takes place to occur via conservative model, two bands are regarding to heavy DNA and another unlabelled DNA should be got. However if replication is semi-conservative, daughter DNAs should possess one labelled strand and one unlabeled strand, thus they will create only one band with intermediate density. The results display that they form bands with intermediate density only in the first generation that ensures the semi conservative model of replication.
