Problem
Overall goal: Appropriate choice of target sites for gene editing is an important part of the process. You'd like to study genes in the fruit fly model system. Look at the following genes we previously studied in FlyBase - Nej, Wg, Gcn5. Use FlyBase and the online tools below to choose a portion of the gene for modification. You will show what sequence you chose and how you would inactivate it with TALENs and CRISPR/Cas. In your design, please indicate any spacers or special added sequences you would use to target your gene.
Instructions:
In FlyBase, type in your gene in the upper right where it says "jump to gene". Note the direction it runs in the Gbrowse browser-type representation. Going back to the previous page, bring up its sequence (in the "sequence" box) using the button for "decorated FASTA". This is a commonly used sequence format. Note that there will be a start codon (ATG) somewhere in that sequence - look at the CDS (coding sequence).
