Specimen must be placed in a sterile vessel, promptly transported to the laboratory to avoid contamination. The granules must be washed with sterile saline repeatedly to eliminate the bacteria. Color, consistency, size, microscopical appearance of granules must be observed.
Direct Examination:
Granules are crushed and inspected in saline and KOH.Staining:
Gram stain:
The granules are compressed, fixed and Gram stained. The filaments are Gram positive. From the size of the filament it is recognized whether it is Eumycotic or Actinomycotic granules.
