The chemical agent is tested against a particular microorganism (i.e., test organism) in Tube- Dilution and Agar-plate methods and the reduction in growth, or complete absence of growth is recorded. The water soluble liquid substances are diluted and dispensed into sterile tubes. A measured amount of test organism is inoculated and at regular intervals, a transfer is made from this tube to other tubes having sterile media.

Such tubes are incubated and the appearance of growth is noticed. By performing a plate count at particular intervals the number of organisms killed per unit time can also be determined by this technique.

The chemical agent can be tested by incorporating it into an agar medium or broth to which the test organism is inoculated. The reduction in the growth or nonattendance of growth shall be assessed after a period of incubation. Broth or agar medium without chemical agent and inoculated with organism will serve up as a standard for comparison. The inhibition by a chemical can be charged by placing the chemical agent in agar medium inoculated with test organism. After incubation the plate is noticed for inhibition zone. Generally a solution of the test chemical is impregnated into absorbent (i.e., filter) paper or poured in hollow cylinders or in agar wells cut by means of a sterile cork borer. Gaseous substances are computed by exposing paper strips impregnated with bacterial spores under particular conditions and then assessing the survivors subsequent to exposure.
