In Oxidative phase, glucose is oxidized and decarboxylated along with the creation of pentose by phosphogluconic acid as displayed in the flow chart. The necessary characteristic of this phase is the production of NADPH2.

1. Glucose is phosphorylated to glucose-6-phosphate through hexokinase.
2. The glucose-6-phosphate is oxidized to 6-phospho-gluconolactonate in the existence of NADP+ through enzyme glucose-6-phosphate dehydrogenase. NADP+ is reduced to NADPH.

3. The 6-phosphogluconolactone is hydrolysed through gluconolactonase to create 6-phosphogluconic acid.

4. The 6-phosphogluconic acid goes through oxidative decarboxylation again in the existence of NADP+ to create Ru5P. This reaction is catalyzed through 6-phosphogluconic dehydrogenase. To NADPH the NADP+ is reduced.

In this reaction CO2 is released.
