Km Value Definition:

Km is described as the concentration of the substrate to which the velocity of the reaction is half maximal. Of enzyme concentration it is independent. The unit of Km is moles per litre. So, Michaelis – Menton constant might be determined though a plot generally known as M-M plot acquired through plotting substrate concentration [S] versus rate of the reaction [V].

Transformation of the Michaelis–Menton equation:

The value of Km might be acquired more correctly from the Lineweaver-Burk equation that is acquired via taking the reciprocal of both sides of the Michaelis–Menton equation.

1/V = Km + [S] / Vmax  [S]                        (1)

Rearranging this equation, we get

1/V  = Km / Vmax [S] + [S] / Vmax [S]                (2)

That is further simplified to

1/V = Km /Vmax . 1/ [S] + 1/ Vmax                (3)
 
A plot of 1/V vs. 1/S (a double reciprocal plot) yields a straight line along with the slope of Km /Vmax and ordinate intercept of 1/ Vmax. Because the slope and intercept are readily calculated from the graph, the Vmax and Km can be correctly determined in the below diagram.
