You are interested in identifying genes that confer resistance to Nocodazole, a microtubule poison that would normally arrest cells in mitosis. You transfect your ORF expression library into your cell line and begin to culture them in Nocodazole. After 1 week, all the cells in your control plate (untransfected cells) are dead. Meanwhile, your transfected condition has colonies that continue to grow.
