How would you determine if they are being transcribed

Problem

You are interested in identifying genes that confer resistance to Nocodazole, a microtubule poison that would normally arrest cells in mitosis. You transfect your ORF expression library into your cell line and begin to culture them in Nocodazole. After 1 week, all the cells in your control plate (untransfected cells) are dead. Meanwhile, your transfected condition has colonies that continue to grow.

1. How would you identify what genes are responsible for Nocodazole resistance in these colonies?

2. How would you determine if they are being transcribed?

3. How would you identify the cellular binding partner(s) of a protein encoded by one of these genes?
