Acetic Acid Test

About 2 - 8 mgm/dl of protein is excreted in urine which is not usually detected by routine test. The testing of urine for protein of which albumin is the main organic constituents is widely used laboratory procedure. Detection of an abnormal amount of protein in urine is a reliable indicator of renal disorder including complications associated with diabetes mellitus.

Take urine in a clean test tube filling 2/3rd portion and boil upper portion of urine for 2 minutes by holding with a test tube holder over a burner flame. A white cloud\ appears in the heated portion if proteins (including albumin) and phosphates are present. Now you add 2 - 3 drops of 5 - 10% glacial acetic acid. If the cloudiness disappears it is due to phosphate, if it persists, then it implies proteinuria (Albuminiuria).
