Assignment, three viruses each

Virus,genbank accession number, sequence of your mutant (containing one or multiple sequence changes, so discuss the effect of the combined mutations not the individual mutations )

What to do?

Go to ncbi site and paste or type your accession number to retrieve your sequence.

You have to do this for three viruses:

Check carefully what is the change made by the mutation(s). Make an educated guess about (=describe) the effect of the mutation(s).

Describe an experiment to establish which step(s) in the life cycle of the virus will be affected by the mutation (e.g. in vitro translation, in vitro replication using purified RdRP, Northern blot, Western blot, in vivo luciferase assay, etc, etc). (Note that a reduced titer does not tell you which step in the life cycle is affected).

Describe also how you make your mutant DNA or RNA.

In some cases the mutation(s) affect more than one function, choose one affected function to test.

You cannot use the same experimental setup for all three viruses in case their mutations are predicted to have a similar effect.

Use the powerpoint presentations, Pubmed (perhaps a similar mutation was already tested), MFOLD, or BLAST, to help you out.

Describe your findings for the three virus mutants in a short report (~half A4 per virus should be sufficient, excl figures and used references)
