Problem

Adenovirus has a complex replication initiation scheme that utilizes the pTP protein as the primer. Replication typically takes place on the linear, double-stranded Adenovirus genome that has the TP protein covalently bound to each 5' end. In the lab, you purify the ds Adenovirus genome, denature it, and isolate one of the single-strands of the genome with the TP protein still attached to the 5' end. If you transfected this single-strand template into permissive, Adenovirus infected cells (to supply all the necessary transacting replication factors) would the single-strand template replicate? Explain why or why not.
