Problem

The following questions deal with renaturation kinetics. The rate at which renaturation occurs can be monitored by cooling a sample in a cuvette while monitoring absorbance at 260 nm.

1. You have one cuvette containing 10 ng/ul and another containing 50 ng/ul of the same DNA sequences. Which one will renature faster? Why?

2. In a different experiment, you have one cuvette containing 150 different DNA sequences and another that contains 1,500 different DNA sequences. Both tubes have the same total DNA concentration and the sequences are 200 bp long. Which one will renature faster? Why?

3. In a third experiment, you have two cuvettes containing 150 different DNA sequences. Both are at the same total DNA concentration, but the sequences in the first tube are 200 bp long while the sequences in the second tube are 2,000 bp long. Which one will renature faster? Why?
