When cloning a gene into an expression vector, people frequently use two different restriction enzymes so that the passenger DNA can be inserted in the correct orientation with respect to the promoter sequence present in the plasmid. But orientation is probably less important when producing an interfering RNA. Why is orientation less important in this case?
