You decide to test the effects of various point mutations on citrate synthase function. Using site-directed mutagenesis, you decide to produce 3 different mutant enzymes: H274K, H274V, and H274D. Discuss why each of the following point mutations was selected and hypothesize the effect it may have on citrate synthase enzymatic function (i.e. substrate binding and mode of catalysis).
