If you were trying to optimize, which three variables would you test first? Why?

I think the first three variable I would test should be kept simple. If the first tests we do are too complex and have multiple outcomes locking in on a sure change could be missed or unworthy of the change. For that I would test these three variables first:
Page graphics: I would test three different layouts on the home screen of the website to test the effectiveness of each. From this I could apply the learned information to other pages to develop a seamless easy to visualize format for the visitors to the page. Just because I think a page looks good and meets the needs does not mean the visitors to the page feel the same.
Timing: I would secondly test the timing of my email marketing efforts to visit rate on the site. This variable could help identify when the best times and modes of delivery can be used to reach the visitors to maximize conversion. This variable would also allow me to setup times that would work best for me to do maintenance on the website. It would be horrible to shut the website down to make changes when the page may be seeing a high rate of visitors searching for the page.
Keyword: The third piece would be identifying the most effective keywords early in the development of the website to optimize its capabilities early. I would try small batches of keywords so it would be easy to identify what is working and what is not working. This variable will help grow organic search capabilities faster than shooting holes in the sky.
