A pharmaceutical chemist wishes to compare two types of reversed-phase columns for the separation and analysis of the drug phenytoin in serum samples. The first columnhas a 3.0 mm innerdiameter and is 6.0 cm. long, containing a C18 stationary phase and 3 um diameter support particles. The second column has an inner diameter of 4.6 mm. and is now having 5 um diameter support particles. Both columns have similar phase ratios and provide the same retention factors for phenytoin in the presence of equivalent mobile phases. (a) At 1.0 mL/min, the first column has a void time of 0.34 min. and gives a retention time of 30.52 min. for phenytoin in solvent A and 10.27 min in solvent B. What is the retention factor for phenytoin in both of these solvents. (b) If the second column has a void time of 1.33 min at 1.0 mL/min, what is the expected retention time for phenytoin on this column in solvents A and B?
