Phosphate group of one nucleotide binds to the pentose of other nucleotide and so forth to generate the polynucleotide chain. Each extremity of the DNA or RNA chain can be distinguished from other extremity consistent with their terminal chemical entity. The phosphate-ended extremity is named as 5’-extremity and pentose-ended extremity is named as 3’-extremity.

Accordingly DNA or RNA chains can be run along with the 5’-3’ way or along with the 3’-5’ way. These ways are significant in various biological actions of DNA and RNA because some of the reactions specifically happen in following one way or the other way.
