Can amine group of peptide bond function as nucleophile


Consider the fully extended conformation of a peptide to answer the following questions. Is there a pattern to the arrangement of the "R" groups, if so, describe it. Even though the carbonyl and amine are connected by a single bond, there is essentially no rotation around this bond, why is that? Can the amine group of the peptide bond function as a nucleophile?
