This method is employed to detect bacterial species in two communities that is, Gram negative and gram positive. This method is depended on physical and chemical properties of their cell walls. It can be employed to identify peptidoglycan that is present in a thick layer in Gram positive bacteria. Purple or blue color submits to the gram positive bacteria. Red color stain submits to the gram negative bacteria. This method is extremely popularly employed in the detection of bacterial organism.
