It is employed to qualitate the antigen. Suitable dilution of the antiserum is integrated in agar gel. Antigen is added to wells cut on surface of gel. As the antigen diffuses in the agar region, correspondence is established and ring of precipitation is formed. The region of precipitin ring is directly proportional to concentration of the antigen. By comparing the area of precipitin with the standard curve obtained through evaluating the precipitin area of recognized concentration of antigen, the concentration of the antigen in given sample can be determined.
