It is employed to qualitate the antigen. Appropriate dilution of antiserum is integrated in the agar gel. Antigen is added up to the wells cut on the surface of gel. Since the antigen diffuses into agar area, equivalence is recognized and ring of precipitation is made. The region of precipitin ring is directly proportional to the concentration of the antigen. By comparing the region of precipitin with a standard curve received by measuring the precipitin region of known concentration of antigen, the concentration of antigen in given sample can be established.
